Am I able to use my Interrail ticket on the route from Brussels (Belgium) to Frankfurt (Germany) ?
Or do I have to pay extra on this route ?

Yes.

I recommend to use the thrice daily direct ICE connections. Do not use the Thalys between Bruxelles and Cologne, it requires an additional reservation which is way to expensive for InterRailers! The ICE does not require a reservation. Since there are only three daily ICE you could also go by a domestic SNCB IC to Liege, then by IR to Aachen, and then with domestic DB trains via Cologne to Frankfurt.
